Immobilization of pyrogallol red with gelatin gel and appliance the composite for total protein determination

Reagent immobilization in condensed media can lead to the considerable change in properties of the
used reagents and improvement of metrological characteristics of analytical methods. Transparent hard
polymeric materials including hardened gelatin gel, which has considerable sorption power, are especially
convenient for analytical purposes.
The objective of this work is investigation of regularities of sorption of pyrogallol red (PGR) in
hardened gelatin gel and evaluation of possibilities of total protein determination in biological fluids with
pyrogallol red, immobilized in gelatin matrix. Behavior of PGR, laws of dye sorption in hardened gelatin gel
and changes in reagent acid properties during it transport from solution to the gelatin gel layer are
investigated.
Condition of reaction between immobilized in gelatin gel PGR and the protein are optimized. The
method of sorption-spectroscopic determination of total protein in biological fluids, which allows expanding
of determinated concentrations and decreasing of total protein detection limit, is developed. Calculated
detection limit of total protein by means of this method is 6·10

g/l. Transparence, mechanical durability, simplicity of making and color
stability (when storing) of solid-phase reagent, make the developed sensor convenient for sorptionspectrometric
and visual-test determination of protein on application for mass analysises.
