Separation of α- and β-alanine, amino acid and dipeptide mixtures by molecular printer polymers
Abstract
The article describes separation of amino acid mixtures, α- and β-alanine, and glycyl glycine. The aim
of the study was to extract α-alanine from mixtures of amino acids of α- and β-alanine, α-alanine and glycine, and glycyl glycine by molecularly imprinted polymers.
A high selective sorbent based on copolymer of 1,2,4,5-benzene tetra-tetracarboxylic acid dianhydide
with 4,4'-diaminodiphenyl oxide was obtained for separation of amino acids. α-alanine was used as a template.
Initial solutions of amino acids with the concentration of 0.5 mol/dm3 were passed through cartridges
filled with 3g of sorbent based on MIPs-α-alanine at the flow rate of 0.2 sm3/min. The volume of solutions was 25 ml. Amino acids were identified in eluate (15 sm3) by liquid chromatography using Agilent 1260 Infinity (Agilent Technologies, CA,US). 0.30±0.02 mol/dm3 α-alanine was detected after the mixture of amino acids and α-alanine with glycyl glycine in eluate had been passed through the cartridge with MIPs-α-alanine. It was established that polymers with molecular imprints of α-alanine only adsorb the target amino acid, α-alanine, and are incapable of molecular recognition of β-alanine, glycine, and glycyl glycine.
The proposed method of separating the structural isomer of alanine and the mixture of amino acids on
molecularly imprinted polymers is very promising as the existing methods of separating amino acid isomers are costly and too specific.
