Abstract

Respiratory nitrate reductase (E.C. 1.7.5.1) from bacterium Thiothrix lacustris AS cultivated under
anaerobic conditions in the presence of nitrate was isolated and purified 78-fold using a multistep purification
scheme including gel-filtration, ion-exchange chromatography, and preparative electrophoresis. The enzyme
was homogenous according to electrophoretic data, with activity of 97,7 U /mg. Kinetic and physicalchemical
characteristics of the enzyme (pH and temperature optima, thermostability, effect of different
inhibitors on activity) were studied.
